General description

ColorWheelColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheelColorWheel® antibody with any ColorWheelColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheelColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheelColorWheel® technology requires both ColorWheelColorWheel® antibody and ColorWheelColorWheel® dye for Flow Cytometry application.
Integrin beta-4 (UniProt: A2A863; also known as CD104) is encoded by the Itgb4 gene (Gene ID: 192897) in murine species. Integrins are heterodimeric cell surface receptors composed of a variable a subunit and a conserved b subunit. The exact combination of various a and b subunits dictate the binding specificity of integrins to different ECM components. Integrins are not constitutively active. Activation of an integrin from low ligand-binding affinity state to high ligand-binding affinity state requires conformational change that can originate from either from their cytoplasmic or extracellular interactions. The recognition site for most integrins that bind the ECM consists of an RGD (arginine-glycine-aspartic acid) sequence. Integrins bind to their ligands with low affinity and this binding occurs only when a certain minimum number of integrins are present at specific points known as focal contacts. Integrin b-4 is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-28), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 29-712), a transmembrane domain (aa 713-733), and a cytoplasmic domain (aa 734-1818). It contains four fibronectin type III domains that are known to bind BPAG1 (Dystonin) and plectin. Integrin b-4 heterodimerizes with a-6 that serves as a receptor for laminin and plays a critical structural role in the hemidesmosome of epithelial cells. It undergoes palmitoylation by DHHC3 at several cysteines of the membrane-proximal region that enhances its stability and surface expression. Palmitoylation also promotes its secondary association with tetraspanins. Integrin b-4 is reported to play a tumorigenic and pro-metastatic role that is mediated by Slug and silencing of Itgb4 gene represses cell proliferation, colony forming ability and cell invasiveness in hepatocellular carcinoma. No. CWDS-PE ColorWheelColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of Cat. No. CWA-1018 Rat IgG2a isotype control and Cat. No. CWDS-PE ColorWheelColorWheel® Antibody-Ready Phycoerythrin (PE) Dye.

Tested Applications

Staining of one million bEnd.3 cells was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1123, Anti-Mouse CD104 (346-11A) ColorWheelColorWheel® Dye-Ready mAb and Cat. No. CWDS-APC ColorWheelColorWheel® Antibody-Ready Allophycocyanin (APC) Dye or an equivalent amount of Cat. No. CWA-1018 Rat IgG2a isotype control and Cat. No. CWDS-APC ColorWheelColorWheel® Antibody-Ready Allophycocyanin (APC) Dye.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Biochem/physiol Actions

Clone 346-11A is a rat monoclonal antibody that detects murine CD104 (Integrin b4). It targets an epitope within the N-terminal, extracellular domain.

Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Preparation Note

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
