General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.
Ectonucleoside triphosphate diphosphohydrolase 1 (UniProt: P49961; also known as EC:3.6.1.5, NTPDase 1, Ecto-ATP diphosphohydrolase 1, Ecto-ATPDase 1, Ecto-ATPase 1, Ecto-apyrase, Lymphoid cell activation antigen, CD39) is encoded by the CD39 (also known as ENTPD1) gene (Gene ID: 953) in human. CD39 is a multi-pass membrane glycoprotein with two cytoplasmic domains (aa 1-16 and 500-510), two transmembrane domains (aa 17-37 and 479-499), and one extracellular domain (aa 37-478). It is expressed primarily on activated lymphoid cells. Its expression is also reported in endothelial cells. It is a membrane-bound enzyme that cleaves ATP and ADP into AMP. AMP is then converted to adenosine by CD73 on the cell surface. This drives a shift from an ATP-driven proinflammatory environment to an anti-inflammatory milieu induced by adenosine. The sequential activity of the CD39/CD73 pathway is important in scavenging extracellular ATP and generate immunosuppressive adenosine in the tumor microenvironment. In the nervous system, it is involved in hydrolysis of ATP and other nucleotides to regulate purinergic neurotransmission. CD39 expression on tumor and endothelial cells is reported to promote angiogenesis and metastatic tumor spread. CD39 overexpression in tumor-bearing mice increases the liver metastasis of MC-26 colorectal tumors in mice. On the other hand, CD39-deficient mice are shown to be resistant to tumor metastases. Mutations in CD39 gene have been linked to an autosomal recessive spastic paraplegia that is characterized by a slow, gradual, progressive weakness and spasticity of the lower limbs. Six isoforms of CD39 have been described that are produced by alternative splicing. (Ref.: Perrot, I., et al. (2019). Cell Rep. 27(8); 2411-2425; Kaczmarek, E., et al. (1996). J. Biol. Biochem/physiol Actions

Clone TU66 is a mouse monoclonal antibody that detects human CD39. It targets an epitope within the extracellular domain.

Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Preparation Note

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
