General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Specificity

Clone TRAIL-R4-01 is a mouse monoclonal antibody that detects CD264/TRAIL-R4. It targets an epitope within the extracellular domain.

Immunogen

Recombinant protein fragment corresponding to amino acids 1-210 of human CD264/TRAIL-R4, hIgGhc fusion protein.

Evaluated by Flow Cytometry in HEK293T cells transiently transfected with human CD 264.

Flow Cytometry Analysis: Staining of one million HEK293T cells transiently transfected with human CD264 was performed using 5 µL of a 1:1 mixture of Cat. No. CWA-1168, Anti-Human CD264 (TRAIL-R4-01) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of Cat. No. CWA-1013 Mouse IgG1 isotype control and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye.

Tested Applications

Flow Cytometry Analysis: Staining of one million HEK293T cells transiently transfected with human CD264 was performed using 5 µL of a 1:1 mixture of Cat. No. CWA-1168, Anti-Human CD264 (TRAIL-R4-01) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-APC ColorWheel® Antibody-Ready Allophycocyanin (APC) Dye or a 1:1 mixture of an equivalent amount of Cat. No. CWA-1013 Mouse IgG1 isotype control and Cat. No. CWDS-APC ColorWheel® Antibody-Ready Allophycocyanin (APC) Dye.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Tumor necrosis factor receptor superfamily member 10D (UniProt: Q9UBN6; also known as Decoy receptor 2, DcR2, TNF-related apoptosis-inducing ligand receptor 4, TRAIL receptor 4, TRAIL-R4, TRAIL receptor with a truncated death domain, CD264) is encoded by the TNFRSF10D (also known as DCR2, TRAILR4, TRUNDD, UNQ251/PRO288) gene (Gene ID: 8793) in human. CD264 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-55), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 56-211), a transmembrane domain (aa 212-232), and a cytoplasmic domain (aa 233-386). It is widely expressed and is observed in fetal kidney, lung, and liver and in adult testis and liver. Its expression is also observed in peripheral blood leukocytes, colon, small intestine, ovary, prostate, and other tissues. CD264 serves as a decoy receptor for tumor necrosis factor-related apoptosis-inducing ligand (TRAIL). It contains a truncated intracellular death domain that prevents it from signaling for TRAIL-induced apoptosis. It inhibits cell death by competing for TRAIL binding and obstructing the formation of a death-inducing signaling complex. CD264 also serves as a surface marker of cellular age for mesenchymal stem cells (MSC). Its levels are upregulated in MSCs at an intermediate stage of cellular aging and remains upregulated as aging progresses towards senescence. (Ref.: Madsen, SD., et al. (2017). Stem Cell Res. Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 µL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 µL PBS. Protect from light.
