General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.
Killer cell lectin-like receptor subfamily B member 1 (UniProt: Q12918; also known as C-type lectin domain family 5 member B, HNKR-P1a, NKR-P1A, Natural killer cell surface protein P1A, CD161) is encoded by the KLRB1 (also known as CLEC5B, NKRP1A) gene (Gene ID: 3820) in human. CD161 is a single-pass, disulfide-linked, homodimeric type II membrane protein serves as a C-type lectin-like receptor. It contains a cytoplasmic domain (aa 1-45), a transmembrane domain (aa 46-66), and an extracellular domain (aa 67-225). It is expressed in a subset of NK cells predominantly in intestinal epithelium and liver. It is also detected in peripheral blood T-cells, including both and β TCR-expressing subsets. During NK cell development, it may facilitate cross-talk of NK cell precursors with cells within the bone marrow. It is reported to play an inhibitory role on natural killer (NK) cells cytotoxicity. Binding of CD161 to its ligand lectin-like transcript 1 (LLT1) is shown to have both inhibitory and costimulatory effects. Its activation results in specific acid sphingomyelinase/SMPD1 stimulation with subsequent marked elevation of intracellular ceramide. Activation also leads to Akt 1 and RSK1 stimulation. No. CWA-1178, Anti-Human CD161/KLRB1 (HP-3G10) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or the equivalent amount of Cat. No. CWA-1013 Mouse IgG1 isotype control and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye. CD161 expression was observed mostly in CD56 positive population.

Tested Applications

Flow Cytometry Analysis: Staining of one million human peripheral blood mononuclear cells (PBMC) lymphocytes was performed with PE conjugated anti-human CD56 and with 5 µL of a 1:1 mixture of Cat. No. CWA-1178, Anti-Human CD161/KLRB1 (HP-3G10) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-APC ColorWheel® Antibody-Ready Allophycocyanin (APC) Dye or the equivalent amount of Cat. No. CWA-1013 Mouse IgG1 isotype control and Cat. No. CWDS-APC ColorWheel® Antibody-Ready Allophycocyanin (APC) Dye. CD161 expression was observed mostly in CD56 positive population. Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone HP-3G10 is a mouse monoclonal antibody that detects CD161/KLRB1. It targets an epitope within the extracellular domain.

Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Preparation Note

1.0 mg/mL after reconstitution at 25 µL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 µL PBS. Protect from light.
