Gene Information

Escherichia coli K12 ... dgkA(948543)

Application

Diacylglycerol Kinase from Escherichia coli has been used in a study to assess the antagonistic regulation of dgkA and plsB genes of phospholipid synthesis by multiple stress responses in Escherichia coli. Diacylglycerol Kinase from Escherichia coli has also been used in a study to identify an alcohol binding site in the first cysteine-rich domain of protein kinase Cdelta.

Biochem/physiol Actions

Specific for the sn-1,2 isomer of DAG. Can be used to measure ceramide production.

Physical form

Supplied as a turbid membrane suspension in 25 mM sodium phosphate buffer, pH 7.0, 20.0% glycerol, 1 mM DTT.

Other Notes

One unit will phosphorylate 1.0 μmol of diacylglycerol per minute at 25°C.

Phospholipid homeostasis of the bacterial membrane is maintained by biochemical regulation of the synthesis enzymes depending on the environment. However, genes encoding phospholipid synthesis enzymes might also be regulated during stress responses, in order for the bacteria to adapt their

The type I diacylglycerol kinase (DGK) isozymes (α, β and γ) contain a shared recoverin homology (RVH) domain, a tandem repeat of Ca2+-binding EF-hand motifs, two cysteine-rich C1 domains, and the catalytic domain. We previously reported that a DGKα mutant

Phosphoinositide cycle plays a pivotal role in neuronal signal transduction. In this cycle, diacylglycerol kinase (DGK) phosphorylates diacylglycerol (DG) to yield phosphatidic acid (PA). DG and PA acts as important second messengers that regulate distinct cascade of cellular events. Previous

We analyzed lipid kinase and lipid phosphatase activities and determined endogenous phytohormone levels by liquid chromatography-tandem mass spectrometry in root and coleoptile tissues following germination of barley (Hordeum vulgare) seeds. The enzymes showing highest activity in aleurone cells were diacylglycerol
