General description

FGFR2 (fibroblast growth factor receptor 2) is a member of the tyrosine kinase receptor family called FGFR. This gene is localized to human chromosome 10q26, and is composed of 2 exons. Its ligand-binding domain in present in its extracellular region, which is composed of three immunoglobulin (Ig)-like domains. It spans the membrane once, and its tyrosine kinase domain is localized to its intracellular region. Alternative splicing of this gene gives rise to two isoforms- epithelial (FGFR2IIIb) and mesenchymal (FGFR2IIIc).

Biochem/physiol Actions

FGFR2 (fibroblast growth factor receptor 2) facilitates tumor growth, invasiveness, metastasis and angiogenesis. It plays an essential role in the development of mammary glands, and its expression is elevated in breast cancers. Studies in North Indian population suggest that polymorphisms in this gene might be linked to susceptibility to breast cancer. It is anti-apoptotic in nature, and suppresses p53-mediated apoptosis, by inducing the elevation of MDM2 levels. Mutations in this gene are linked to mild Crouzon syndrome and craniosynostosis.
