General description

IgM antibodies are present as pentamers in the serum and are produced in response to antigens
Anti-Human IgM (μ-chain specific)-FITC antibody specifically binds to mu-chain of human IgM. After immunospecific purification, anti-human IgM is conjugated to fluorescein isothiocyanate (FITC), in an alkaline reaction. The antibody preparation is specific for human IgM when tested against purified human IgA, IgG, IgM, Bence Jones κ and λ myeloma proteins. The product is also determined to have no cross reactivity with mouse or rat serum proteins.

Immunogen

Purified human IgM

Application

Anti-Human IgM (μ-chain specific)-FITC antibody may be used for immunofluorescence at a working antibody dilution of 1:64 using human peripheral blood lymphocytes. A dilution of 1:32-1:64 may be used for anti-nuclear antibody (A.N.A.) assay on acetone-fixed human Hep2 cells. A dilution of 1:32 may be used on acetone-fixed mouse liver cells for anti-nuclear antibody assay. It may also be used for agar diffusion assay.

Biochem/physiol Actions

No cross-reaction with mouse and rat IgG.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Previous studies have discerned two forms of polymeric mouse IgM: moderately cytolytic (complement-activating) pentamer, which contains J chain, and highly cytolytic hexamer, which lacks J chain. To investigate the relationships among polymeric structure, J chain content, and cytolytic activity, we
