Biochem/physiol Actions
A key protein in signal transduction, Grb2 transfers the signal from tyrosine kinases such as epidermal growth factor (EGF) receptor to the small G protein Ras. GRB2 contains an SH2 (Src homology 2) domain which enables binding to tyrosine-phosphorylated receptors. The fusion protein enables detection and purification of tyrosine-phosphorylated proteins (i.e., tyrosine-phosphorylated EGF receptor), utilizing the affinity of protein A for the Fc domain of IgG.

A Drosophila gene (drk) encodes a widely expressed protein with a single SH2 domain and two flanking SH3 domains, which is homologous to the Sem-5 protein of C. elegans and mammalian GRB2. Genetic analysis suggests that drk function is essential
