Biochem/physiol Actions
GAP-43 is an intracellular phosphoprotein that is present exclusively in the peripheral and central nervous systems. Protein kinase C (PKC) phosphorylates GAP-43 on a single site, serine 41. This phosphorylated protein is found in the areas where growth cones make productive and stable contacts with other cells. It facilitates stabilization of long actin filaments. Phosphorylation by PKC at serine 41 is related to nerve-terminal sprouting and long-term potentiation as high levels of GAP-43 are present in neocortical areas and in the limbic system. It is crucial in mediating experience dependent plasticity.
Physical form
Solution in 10 mM HEPES, pH 7.5, containing 150 mM NaCl, 100 μg/mL BSA, and 50% glycerol.
Analysis Note
An additional band at 150-160 kDa may be detected and reflects the oligomeric nature of GAP-43.
