Gene Information

human ... CRIPT(9419)

General description

CRIPT (cysteine-rich PDZ-binding protein) is a cytoplasmic, post-synaptic protein. It is highly conserved across mammals and plants. It binds to PDZ3 domain of PSD-95 (postsynaptic density protein 95) throught its C-terminal.

Immunogen

Immunogen Sequence: GI # 7661798, sequence 1-101
Recombinant postsynaptic protein CRIPT

Application

Anti-CRIPT antibody produced in chicken is suitable for western blotting analysis at a dilution of 1:500, for tissue or cell staining at a dilution of 1:200.

Biochem/physiol Actions

Cysteine-rich PDZ-binding protein binds directly to microtubules, and links PSD-95 to the microtubule cytoskeleton. CRIPT, in brain, colocalizes with PSD-95 in the postsynaptic density and can be coimmunoprecipitated with PSD-95 and tubulin. It may help in regulating PSD-95 interaction with a tubulin-based cytoskeleton in excitatory synapses.

Physical form

Solution in phosphate buffered saline containing 0.02% sodium azide.

The thermodynamic parameters associated with the binding of several series of linear peptides to the third PDZ domain (PDZ3) of the postsynaptic density 95 protein (PSD-95) have been measured using isothermal titration calorimetry (ITC). Two strategies were pursued in developing

CRIPT is a postsynaptic protein that binds selectively to the third PDZ domain (PDZ3) of PSD-95. Here we show that CRIPT also binds directly to microtubules, thereby linking PSD-95 to the microtubule cytoskeleton. Disrupting the CRIPT-PSD-95 interaction in cultured hippocampal

The synaptic protein PSD-95/SAP90 binds to and clusters a variety of membrane proteins via its two N-terminal PDZ domains. We report a novel protein, CRIPT, which is highly conserved from mammals to plants and binds selectively to the third PDZ
