Biochem/physiol Actions
Cyclin-dependent kinase 4 (also known as cell division protein kinase 4) is an enzyme encoded by the CDK4 gene in humans. It is highly expressed in nasopharyngeal carcinomas (NPC). CDK4 phosphorylates and inactivates retinoblastoma (Rb) protein family members in association with cyclin-D. It mediates progression through the G1- to the S-phase of the cell cycle. Over expression of CDK4 is mostly independent with gene amplification and is a potential prognostic biomarker in NPC and may indicate tumor aggressiveness through cell cycle dysregulation. It is the main molecular feature of well-differentiated liposarcomas (WDLS) and dedifferentiated liposarcomas (DDLS). Inhibition of CDK4 partly restores G(1)-S arrest and sensitizes cells to doxorubicin-mediated cell death in MYCN-amplified cells with an intact p53 pathway. CDK4 up-regulation takes place in oral cancer progression.

Relapse with drug-resistant disease is the main cause of death in MYCN-amplified neuroblastoma patients. MYCN-amplified neuroblastoma cells in vitro are characterized by a failure to arrest at the G(1)-S checkpoint after irradiation- or drug-induced DNA damage. We show that several

While aberrant expression of cyclin-dependent kinase-4 (CDK4) has been found in squamous cell carcinoma of the head and neck (SCCHN), the associations between CDK4 and its regulators, namely, cyclin D1, cyclin E, gankyrin, SEI1, and BMI1 in gene expression remain
