General description
GRB2 (growth factor receptor-bound protein 2) is an adaptor protein, which is expressed ubiquitously. It is composed of two SH3 domains and one SH2 domain. The SH3 domains are present at the N- and C-termini, and are intervened by SH2 domain. It exists as a heterodimeric protein.

Biochem/physiol Actions
Growth factor receptor-bound protein 2 (GRB2) is involved in signal transduction/cell communication. GRB2 controls fibroblast growth factor receptor 2 (FGFR2) signaling by regulating receptor kinase and SH2 domain-containing protein tyrosine phosphatase 2 (Shp2) phosphatase activities in the absence of extracellular stimulation. The protein is involved in the activation of the Ras signaling pathway. It helps in the regulation of Receptor tyrosine kinases (RTKs) signaling. GRB2 is associated with tyrosine-phosphorylated epidermal growth factor receptors (EGFRs) and platelet-derived growth factor receptors (PDGFRs) through its SH2 domain. It plays a crucial role in a highly conserved mechanism for growth factor control of ras signaling.
