Gene Information
human ... GGH(8836)
General description
GGH (γ-glutamyl hydrolase) gene is composed of nine exons, and is 24kb in size. It encodes a peptidase which is localized to lysosomes. It is a homodimer composed of eleven α-helices, and fourteen β-strands. Its central region contains three and five α-helices sandwiching eight stranded β-sheets.
Application
Anti-GGH antibody produced in chicken is suitable for western blotting analysis at a dilution of 1:500, for tissue or cell staining at a dilution of 1:200.
Biochem/physiol Actions
γ-glutamyl hydrolase plays an important role in the metabolism of folic acid and the pharmacology of antifolates (such as methotrexate). GGH may play a relevant role in the development and progression of invasive breast cancer and may act as an attractive targeted therapy. Polymorphism in the gene is associated with efficacy and side effects of methotrexate for treatment of acute lymphoblastic leukemia (ALL).
Physical form
Solution in phosphate buffered saline containing 0.02% sodium azide.

gamma-Glutamyl hydrolase catalyzes the cleavage of the gamma-glutamyl chain of folylpoly-gamma-glutamyl substrates and is a central enzyme in folyl and antifolyl poly-gamma-glutamate metabolism. The crystal structure of human gamma-glutamyl hydrolase, determined at 1.6-A resolution, reveals that the protein is a
