Gene Information

human ... SLC12A5(57468)

General description

KCCs or K+/Cl- cotransporters belong to the cation-chloride cotransporter (CCC) family, and have four isoforms ranging from KCC1- KCC4. KCC2 or K+/Cl- cotransporter type 2 is one of the main transporters responsible for removing Cl- from neurons. It is encoded by the gene SLC12A5, which is located on the chromosomal region 20q13.12. The expression of KCC2 is exclusive to neurons.

Immunogen

Solute carrier family 12 member 5 recombinant protein epitope signature tag (PrEST)

Application

Anti-KCC2 antibody is suitable for coimmunoprecipitation.
Anti-SLC12A5 antibody produced in rabbit, a Prestige Antibody, is developed and validated by the Human Protein Atlas (HPA) project . Each antibody is tested by immunohistochemistry against hundreds of normal and disease tissues. These images can be viewed on the Human Protein Atlas (HPA) site by clicking on the Image Gallery link. The antibodies are also tested using immunofluorescence and western blotting. To view these protocols and other useful information about Prestige Antibodies and the HPA, visit sigma.com/prestige.

Biochem/physiol Actions

KCC2 (K+/Cl- cotransporter 2) mediates the inhibitory responses of neurotransmitters GABA and glycine by maintaining low intracellular Cl- levels, which results in hyperpolarising postsynaptic responses. KCC2 is essential for maintaining normal motor neuron functions and spinal cord reflexes in neonates. Therefore, any aberrations lead to abnormal motor control, muscle tone and even perinatal death because of lack of ability to breathe. Neuronal plasticity is dependent upon changes in the efficacy of synaptic inhibition, which in turn is mediated by the changes in the expression of KCC2. Human idiopathic generalized epilepsy has been shown to be associated with two KCC2 variants- R952H and R1049C, which have significantly less Cl- extrusion capabilities. KCC2 is also implicated in schizophrenia, where the expression of KCC2 is decreased in the hippocampus of schizophrenic adult patients as compared to the control population.

Features and Benefits

Prestige Antibodies® are highly characterized and extensively validated antibodies with the added benefit of all available characterization data for each target being accessible via the Human Protein Atlas portal linked just below the product name at the top of this page. The uniqueness and low cross-reactivity of the Prestige Antibodies® to other proteins are due to a thorough selection of antigen regions, affinity purification, and stringent selection. Prestige antigen controls are available for every corresponding Prestige Antibody and can be found in the linkage section.

Every Prestige Antibody is tested in the following ways:
  • IHC tissue array of 44 normal human tissues and 20 of the most common cancer type tissues.
  • Protein array of 364 human recombinant protein fragments.

Physical form

Solution in phosphate-buffered saline, pH 7.2, containing 40% glycerol and 0.02% sodium azide
