General description

IFI6 (interferon, α-inducible protein 6) is a part of ISG12 (IFN stimulated gene) family. In humans, there are four members in this family, where IFI6 is located at chromosome 1p35, and the other three at 14q32. It is a hydrophobic protein with a molecular weight of 12.9kDa. It has a conserved ISG12 motif, which is made of ~80 amino acids. When expressed ectopically, IFI6 is found to be localized to mitochondria.

Application

Biochem/physiol Actions

The functional characteristics of IFI6 (interferon, α-inducible protein 6) is not yet well known. This protein is induced by interferons (IFN), and is responsible for cell survival. It is strongly induced by IFN-α2b, where it prevents apoptosis and maintains mitochondrial stability. This way, it also antagonizes the apoptotic effect of TRAIL, especially in tumor cells. Thus, it can be a potential therapeutic target for various malignancies, such as myeloma. IFI6 is also induced by estrogen, and is highly expressed in the initial stages of breast cancer. It alters the balance between pro- and anti-apoptotic members of Bcl-2 family. Hence, it plays a role in poor prognosis of breast cancer, hyperplasia and tamoxifen resistance.
