Gene Information
human ... FGD5(152273)
General description
The gene FGD5 (FYVE, RhoGEF and PH domain-containing protein 5) is mapped to human chromosome 3p25.1. It belongs to FGD family of guanine nucleotide exchange factors. In human umbilical vein endothelial cells, FGD5 is present at peripheral membrane ruffles and perinuclear regions. The protein contains a glutamate-rich amino-terminal region, a Dbl-homology (DH) domain, a first pleckstrin homology (PH) domain, an FYVE (Fab 1, YOTB, Vac 1 and EEA1) domain and a second carboxyl-terminal PH domain.
Immunogen
FYVE, RhoGEF and PH domain-containing protein 5 recombinant protein epitope signature tag (PrEST)
Application
Anti-FGD5 antibody produced in rabbit, a Prestige Antibody, is developed and validated by the Human Protein Atlas (HPA) project . Each antibody is tested by immunohistochemistry against hundreds of normal and disease tissues. These images can be viewed on the Human Protein Atlas (HPA) site by clicking on the Image Gallery link. The antibodies are also tested using immunofluorescence and western blotting. To view these protocols and other useful information about Prestige Antibodies and the HPA, visit sigma.com/prestige. The antibody has been used for immunocytochemistry in monolayer-cultured HUVECs (human umbilical vein endothelial cells).
Applications in which this antibody has been used successfully, and the associated peer-reviewed papers, are given below.

Biochem/physiol Actions
Increase in levels of FGD5 (FYVE, RhoGEF and PH domain-containing protein 5) increases CDC42 (cell division cycle 42) activity. In vascular endothelial cells, FGD5 controls the proangiogenic action of VEGF (vascular endothelial growth factor). It modulates phosphatidylinositol 3-kinase signaling and thereby controls endothelial adhesion, survival and angiogenesis. FGD5 also participates in endothelial cell-targeted apoptosis and regulates vascular pruning.
Features and Benefits
Prestige Antibodies® are highly characterized and extensively validated antibodies with the added benefit of all available characterization data for each target being accessible via the Human Protein Atlas portal linked just below the product name at the top of this page. The uniqueness and low cross-reactivity of the Prestige Antibodies® to other proteins are due to a thorough selection of antigen regions, affinity purification, and stringent selection. Prestige antigen controls are available for every corresponding Prestige Antibody and can be found in the linkage section.
Every Prestige Antibody is tested in the following ways:
Every Prestige Antibody is tested in the following ways:
- IHC tissue array of 44 normal human tissues and 20 of the most common cancer type tissues.
- Protein array of 364 human recombinant protein fragments.
