General description

Cathepsin S (CTSS; EC 3.4.22.27) is a lysosomal cysteine proteinase involved in the degradation of proteins to peptides required for the presentation of MHC class II molecules.1 It is associated with pathological conditions such as atherosclerosis, coronary heart disease, cancer, and insulin resistance.

Biochem/physiol Actions

The Cathepsin S Inhibitor Screening Kit is a simple assay for screening potential inhibitors of CTSS. CTSS activity is measured by cleaving a synthetic 7-amino-4-trifluoromethylcoumarin (AFC)-based substrate to yield AFC, a fluorescent product (λex = 400/ λem = 505 nm), proportional to the enzymatic activity present.
