General description

The new Alkaline Phosphatase (ALP) Assay Kit, MAK530, is now available! Alkaline phosphatase (ALP) catalyzes the hydrolysis of phosphate esters in alkaline buffer and produces an organic radical and inorganic phosphate. The change in alkaline phosphatase level and activity is associated with several diseases in the liver and bones. Alkaline phosphatase is also a popular enzyme conjugated to secondary antibody in ELISA assays.

Application


  • Fibroblast growth factor-4 maintains cellular viability while enhancing osteogenic differentiation of stem cell spheroids in part by regulating RUNX2 and BGLAP expression.: This study used the Alkaline Phosphatase Activity Fluorometric Assay Kit to measure osteogenic differentiation in stem cell spheroids. The results demonstrated that fibroblast growth factor-4 (FGF-4) enhances the expression of osteogenic markers RUNX2 and BGLAP, indicating improved cellular viability and differentiation (Son et al., 2020) (Son et al., 2020).

  • Osteogenic Effect of Inducible Nitric Oxide Synthase (iNOS)-Loaded Mineralized Nanoparticles on Embryonic Stem Cells.: This research employed the Alkaline Phosphatase Activity Fluorometric Assay Kit to assess the osteogenic differentiation of embryonic stem cells treated with iNOS-loaded nanoparticles. The findings revealed significant enhancement in osteogenic activity, suggesting potential applications in regenerative medicine (Lee et al., 2018) (Lee et al., 2018).

  • Calcium Channel Opening Rather than the Release of ATP Causes the Apoptosis of Osteoblasts Induced by Overloaded Mechanical Stimulation.: The study used the Alkaline Phosphatase Activity Fluorometric Assay Kit to investigate osteoblast apoptosis under mechanical stress. It was found that calcium channel opening is the primary cause of apoptosis, providing insights into bone health and disease mechanisms (Liu et al., 2017) (Liu et al., 2017).

The kit is suitable for the measurement of alkaline phosphatase activity in serum, plasma, urine, semen, cell culture media, cells and tissue.

Biochem/physiol Actions

In the Alkaline Phosphatase Activity Fluorometric Assay Kit, ALP cleaves the phosphate group of the non-fluorescent 4-Methylumbelliferyl phosphate disodium salt (MUP) substrate resulting in an intense fluorescent signal (λEx = 360 nm/λEm = 440 nm).
