General description

Nuclear transport factor 2 (NTF2 or NUTF2) is a cytosolic soluble protein. The polypeptide chain of NTF2 has α and β sheets and a hydrophobic cavity. NUTF2 is localized on the human chromosome on 16q22.1.

Immunogen

recombinant human NTF2. The epitope lies within the extreme C-terminus of NTF2.

Application

Monoclonal Anti-NTF2 antibody produced in mouse has been used in western blotting.

Biochem/physiol Actions

Nuclear transport factor 2 (NTF2 or NUTF2) binds with Ras (Rat Sarcoma)-family GTPase (guanosine triphosphatase) Ran (Ras related nuclear protein) and facilitates the transport of Ran to the nucleus. NTF2 binds to nuclear pore complex (NPC) nucleoporin proteins with FxFG repeats especially p62 also binds preferably to GDP-Ran rather than GTP-Ran to form the complex GDP-Ran and p62 binds at different regions in the hydrophobic cavity of NTF2. The mutation in NTF2 hinders the binding and transport of Ran and formation the p62-NTF2-Ran complex.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Analysis Note

The antibody may also be used for microinjection, in vitro nuclear protein import assays, and blocking of NTF2 binding to Ran.
