Biochem/physiol Actions

p53 binding protein 1 (53BP1) is a transcriptional activator of tumour suppressor protein p53. 53BP1 senses the double strand DNA breaks induced by ionising radiations and chemicals and involved in the DNA repair pathway. 53BP1 is a downstream target of ataxia telangiectasia mutated (ATM) phosphorylation. 53BP1 binds to histone 3 lysine 79 methylated residues (H3K79) or dimethylated histone 4 lysine 20 (H4K20me2) via the tandem tudor domain, which enables its interaction with the double strand DNA breaks.

General description

p53 binding protein 1 (53BP1), also known as, tumor protein p53 binding protein 1, is homologous to the DNA damage checkpoint protein Rad9p of Saccharomyces cerevisiae. In the human chromosome, 53BP1 is localized on 15q15.3.

Immunogen

The epitope recognized by PLA0303 maps to a region between residue 350 and 400 of human Tumor Protein p53 Binding Protein, 1 using the numbering given in entry NP_005648.1 (GeneID 7158).

Other Notes

53BP1 (p53 Binding Protein 1) is an early participant in the cellular response to DNA double-strand breaks. 53BP1 functions in a redundant role in phosphorylation of the downstream checkpoint effector proteins Brca1 and Chk2 but was required for the formation of Brca1 foci in a hierarchical branched pathway for the recruitment of repair and signaling proteins to sites of DNA damage.
