General description

Raf-1 is a serine/threonine kinase that is activated by tyrosine kinases . Raf-1 inhibits Ras signaling and subsequently modulates cell proliferation and development . c-Raf expression has been linked to the pathogenesis of lung carcinoma . Anti-phospho-c-Raf pTyr340/341 antibody is specific for human c-Raf that is phosphorylated on tyrosines 340 and 341. Although the product has not been tested for cross-reactivity, the antibody is expected to recognize mouse and rat c-Raf (100% homologous).

Immunogen

synthetic phosphopeptide derived from a region of human Raf that contains tyrosines 340 and 341.

Application

Anti-phospho-c-Raf pTyr340/341 antibody is suitable for use in western blot (1:1,000 using HEK293 cells overexpressing c-Raf).

Physical form

Solution in Dulbecco′s phosphate buffered saline (without Mg2+ and Ca2+), pH 7.3, with 1.0 mg/mL BSA (IgG and protease free), 50% glycerol, and 0.05% sodium azide.

By binding to serine-phosphorylated proteins, 14-3-3 proteins function as effectors of serine phosphorylation. The exact mechanism of their action is, however, still largely unknown. Here we demonstrate a requirement for 14-3-3 for Raf-1 kinase activity and phosphorylation. Expression of dominant

Raf-1 is a key protein involved in the transmission of developmental and proliferative signals generated by receptor and nonreceptor tyrosine kinases. Biochemical and genetic studies have demonstrated that Raf-1 functions downstream of activated tyrosine kinases and Ras and upstream of

The serine/threonine kinase activity of the Raf-1 proto-oncogene product is stimulated by the activation of many tyrosine kinases, including growth factor receptors and pp60v-src. Recent studies of growth factor signal transduction pathways demonstrate that Raf-1 functions downstream of activated tyrosine

We have investigated the role of individual members of the Raf/Mek/Erk cascade in the onset of K-Ras oncogene-driven non-small cell lung carcinoma (NSCLC). Ablation of Erk1 or Erk2 in K-Ras oncogene-expressing lung cells had no significant effect due to compensatory
