General description

Monoclonal Anti-RAP1 A/B (mouse IgM isotype) is derived from the hybridoma R116 produced by the fusion of mouse myeloma cells (NS1 cells) and splenocytes from BALB/c mice. Ras-related protein 1A (RAP1A/Krev-1) belongs to the Ras superfamily of small GTPases. RAP1 gene is mapped to human chromosome 1p13.3

Biochem/physiol Actions

Monoclonal Anti-RAP1 A/B recognizes human and hamster RAP1 A/B (~21 kDa).
Ras-related protein 1A (RAP1A), a novel oncoprotein in human malignancies acts as an important modulator in T cell response. RAP1A also participates in bacterial infection, cell dynamics and osteoblast differentiation. It may serve as a Ras antagonist and is a tumor suppressor gene. RAP1A is activated by growth factors, neurotransmitters and cytokines. Several second messengers, such as cyclic adenosine monophosphate (cAMP), diacylglycerol and calcium may serve as the mediators of this activation. In many cells, active RAP1 changes its cellular location from endosomal membranes to the plasma membrane. RAP1 binds competitively to RAS-GTPase activating protein (RASA) thus interfering with its RAS effectors functions. Ectopically expressed RAP1 is capable of interacting with the RAS effector (RAF1), which may lead to the inhibition of extra-cellular-signal-regulated kinase (ERK) activation. RAP1 can also mediate cyclic adenosine monophosphate (cAMP)-induced ERK activation.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, and 15 mM sodium azide.

Preparation Note

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
