General description

The activation of sphingosine kinase plays a role in the pathogenesis of transplant arteriosclerosis.

Application

Sphingosine kinase 2 has been used in a study to assess novel therapeutic targets for rheumatoid arthritis. Sphingosine kinase 2 has also been used in a study to determine that crowding induces live cell extrusion, which maintains homeostatic cell numbers in epithelia .

Biochem/physiol Actions

Converts sphingosine to sphingosine-1-phosphate. SPHK2 shares two large conserved regions with SPHK1. While SPHK1 is found only in the cytosol, SPHK2 is localized in both the cytosol and nuclei.

Physical form

Supplied at 1.0 mg/ml in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween 20, 10% glycerol, 3 mM DTT.

Other Notes

One unit will produce 1.0 pmol of sphingosine-1-phosphate per minute at pH 7.5, 25 °C.

Rheumatoid arthritis (RA) is a chronic, destructive, autoimmune joint disease characterized by elevated levels of proinflammatory cytokine production. Sphingosine kinase (SphK) phosphorylates sphingosine into sphingosine-1-phosphate. Synovial fluid of RA patients exhibits significantly higher levels of S1P than their non-inflammatory osteoarthritis

Transplant arteriosclerosis is a major cause of late intestinal allograft dysfunction. However, little is known about the immunologic and molecular mechanisms underlying it, and no effective treatment is available. This study aimed to investigate the role of sphingosine kinase 1

For an epithelium to provide a protective barrier, it must maintain homeostatic cell numbers by matching the number of dividing cells with the number of dying cells. Although compensatory cell division can be triggered by dying cells, it is unknown
