General description
Synemin (SYNM), also known as desmuslin/DMN is a unique type VI intermediate filament (IF) protein. The synemin gene codes for three isoforms, such as a-synemin, b-synemin and L-synemin with overlapping distribution. The three isoforms share the same head and rod domains but differ at their C-terminus. These isoforms are expressed in muscle and brain, in astrocytes, glia and neuronal cells and in the vascular system. In human skeletal myofibers, b-synemin is located to the high stress regions. SYNM gene is located on human chromosome 15q26.3.
Application
Anti-Synemin antibody produced in rabbit may be used in immunoblotting.
Biochem/physiol Actions
Anti-Synemin specifically recognizes rat synemin.
Synemin protein interacts with major intermediate filament (IF) proteins including desmin, vimentin and glial fibrillary acidic protein (GFAP) and is thought to play an important role in maintaining structural integrity of the cell. In astrocytoma with migratory properties, synemin contributes to the actin cytoskeleton dynamics. This protein play a major role as a bridging protein between IFs and focal adhesions in rat and human liver tissues. Synemin may also participate in cytoskeletal cross?linking. This IF?associated protein is also known to play a key role in human muscle diseases.
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note
Store at –20 °C. For continuous use, the product may be stored at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
