Biochem/physiol Actions
Cereblon (CRBN) is a substrate binding component of the E3 ubiquitin ligase complex. It interacts with thalidomide and its analog to facilitate proteasome-dependent degradation of the transcription factors, Ikaros family zinc finger protein 1 and 3 (IKZF1 and IKZF3). CRBN provides cytoprotection against the ubiquitin-proteasome system-dysfunction. It negatively regulates the AMP-activated protein kinase signaling pathway in vivo. Nonsense mutation of the CRBN gene causes mild type of autosomal recessive nonsyndromic intellectual disability (ID).
