General description

The eukaryotic translation initiation factor 2C, subunit 2 (EIF2C2) or protein argonaute-2 (Ago2) belongs to the argonaute protein subfamily. The argonaute proteins contains the piwi-argonaute-zwille (PAZ), the P-element induced wimpy testis (PIWI) domain and the MID domain. Ago2 is localized to the processing bodies (P bodies) and the nucleus. The AGO2 gene is mapped to human chromosome 8q24.3.

Immunogen

synthetic peptide corresponding to the N-terminal region of human EIF2C2/Ago2 conjugated to KLH. The corresponding sequence differs by one amino acid in rat and mouse.

Application

Anti-EIF2C2/Ago2 (N-terminal) antibody produced in rabbit is suitable for immunoblotting.

Biochem/physiol Actions

Anti-EIF2C2/Ago2 recognizes human EIF2C2/Ago2.
The protein argonaute-2 (Ago2) is considered as a catalytic unit of mammalian RNA interference (RNAi). It possesses a slicer endonuclease activity that cleaves the target RNA in RNAi. The mRNA cleavage is possible by ribonucleoprotein complexes. Ago2 mediates tissue-specific developmental processes. Ago2 regulates both, the transcriptional and posttranslational modifications. It is implicated in the tumorigenic progression of breast cancer cell lines.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Preparation Note

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
