General description
The mammalian lethal with SEC13 protein 8 (MLST8) gene is mapped to human chromosome 16p13.3. mLST8 also known as GβL (G protein β-subunit-like protein) consists of seven WD40 (tryptophan-aspartic acid/ β-transducin) repeats and has similar sequence homology with the β subunits of heterotrimeric G proteins. mLST8 is a subunit of the mammalian target of rapamycin (mTOR) signaling complexes, mTORC1 and mTORC2. mLST8 is conserved among all eukaryotes and is a widely expressed protein.
Immunogen
synthetic peptide corresponding to a sequence located near the C-terminus of human mLST8, conjugated to KLH. The corresponding sequence is identical in mouse, rat, canine, bovine and monkey
Application
Anti-mLST8/GBL (C-terminal) antibody produced in rabbit is suitable for immunoblotting and immunoprecipitation.
Biochem/physiol Actions
Anti-mLST8/GBL (C-terminal) recognizes human mLST8.
Mammalian lethal with SEC13 protein 8 (mLST8) stimulates the mammalian target of rapamycin (mTOR) kinase function by associating with the kinase domain of mTOR. mLST8 is shown to be a critical molecule in the nutrient-sensitive raptor-mTOR interaction. This protein might also be involved in the raptor-mediated inhibition of mTOR kinase activity. mLST8 is known to mediate the phosphorylation of S6 Kinase 1 (S6K1), in vivo. This infers the role of mLST8 in positively regulating the mTOR pathway. Overexpression of the mLST8 gene has been observed in hepatocellular carcinoma (HCC).
Physical form
Solution in 0.01 M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.
Preparation Note
For continuous use, store at 2-8°C for up to one month. For extended storage freeze in working aliquots. Repeated freezing and thawing is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.

mTOR and raptor are components of a signaling pathway that regulates mammalian cell growth in response to nutrients and growth factors. Here, we identify a member of this pathway, a protein named GbetaL that binds to the kinase domain of

The mechanistic target of rapamycin (mTOR) pathway, containing mTOR complex 1 (mTORC1) and mTORC2, is dysregulated in multiple cancers, including hepatocellular carcinoma (HCC). Mammalian lethal with sec-13 protein 8 (mLST8) is a shared constituent of both mTORC1 and mTORC2, yet
