Biochem/physiol Actions

AA147 is an ATF6α (ATF6) pathway-selective ER proteostasis network (PN) regulator (63.3% ATF6 vs.17.5% IRE1 activation, respectively, by HEK293-based ERSE- or XBP1s-luciferase reporter assay; 6.8 μM, 18 h) that selectively activates ATF6 target transcripts production over those of XBP1s & PERK (10 μM; HEK293T-Rex cells), inducing BiP mRNA in HepG2 and wt, but not Atf6-/-, MEFs (10 μM). AA147 enhances degradation and ER retention of destablized amyloidogenic transthyretin TTR(A25T) and Ig LC, respectively, exogenously expressed in HepG2 and ALMC-2. ATF6 activation by AA147 is also reported to suppress hESCs pluripotency and promotes mesodermal differentiation.
ATF6α-selective activator that reduces the secretion of destablized amyloidogenic proteins and promotes hESCs mesodermal differentiation.
