Biochem/physiol Actions

HS-1371 is a potent ATP-competitive RIP3 (RIPK3) kinase inhibitor (IC50 = 20.8 nM, [ATP] = 10 μM) that blocks RIP3 S227 autophosphorylation and selectively suppresses TNF/SMAC mimetic/Z-VAD-FMK agonist treatment-induced necroptosis induction in HT-29 cultures (by 44.4% at 1 μM, by 77.7% at 5 μM) in a RIP3- and MLKL-dependent manner.
