Inhibitor of phosphatidylinositol 4-phosphate 5 kinase type 1C (PIP5K1C).
UNC3230 is a selective inhibitor of the lipid kinase phosphatidylinositol 4-phosphate 5 kinase type 1C (PIP5K1C) with an IC50 of 41 nM. UNC3230 showed selectivity for PIP5K1C and PIP4K2C (phosphatidylinositol 5-phosphate 4-kinase, type II, gamma) and did not inhibit PIP5K1A or any of the other lipid kinases that regulate phosphoinositide levels, including phosphatidylinositol 4,5-bisphosphate 3-kinases (PIK3s, also known as PI3Ks). UNC3230 lowered PIP2 levels in nociceptive dorsal root ganglia (DRG) neurons and had antinociceptive effects when injected intrathecally as well as peripherally into the hindpaw, indicating that either central or peripheral inhibition of PIP5K1C can reduce nociceptive responses.
