SARS-CoV and SARS-CoV-2 main protease (Mpro; 3CLpro) fluorescence energy transfer (FRET) substrate with >10-fold sensitivity than Edans-Dabcyl-based substrate.

Abz-SVTLQSG-TyrNO2R (Abz = 2-aminobenzoyl; TyrNO2 = 2-nitro-L-tyrosyl) is a substrate for the detection of SARS-CoV and SARS-CoV-2 main protease (Mpro; 3CLpro) activity by fluorescence energy transfer (FRET) with >10-fold sensitivity than the corresponding Edans-Dabcyl-based FRET peptide. Substrate cleavage by Mpro between Gln and Ser leads to increased fluorescence (ex 330+/-10 nm, em 420+/-20 nm) as a result of the separation of the N-terminal Abz (fluorescent donor) and the C-terminal TyrNO2 (fluorescent quencher).
