General description
Maleimide-PEG2-biotin, a maleimide-conjugated biotin molecule with a medium-length spacer arm (PEG)2, is membrane-impermeable and exhibits high affinity for free thiol groups on cysteines.
Application
Maleimide-PEG2-biotin is suitable for use in labeling and purifying biomolecules with high specificity. It may be used in mass spectrometry to detect free sulfhydryl residues in intact proteins.
Biochem/physiol Actions
Biotin-PEG2-Mal, a biotin maleimide linker, forms stable thioether bonds by covalently labeling cysteine sulfhydryl groups on antibodies or proteins and can be detected via streptavidin-HRP in Western blot assays following pull-down. Maleimide labels react with sulfhydryl (–SH) groups at acidic pH, minimizing the risk of disulfide bond scrambling during labeling that can happen at higher pH levels.
