General description
Very-low-density-lipoprotein receptor (VLDLR) is a 130 kDa type I transmembrane protein in the low density lipoprotein (LDL) receptor family that plays a significant role in lipid metabolism and in nervous system development and function. The VLDL receptor structure is more or less homologous to that LDL receptor, excluding the eightfold repeated ligand binding domain of VLDLR, which is sevenfold in LDLR. The protein sequence of VLDLR is mostly conserved among mammals such as rabbit, human and mouse. The gene is mapped to human chromosome 9p24.2.
Biochem/physiol Actions
Very-low-density-lipoprotein receptor (VLDLR) is associated with the metabolism of triglyceride rich lipoproteins (VLDL,β-migrating VLDL and intermediate-density lipoprotein) involving apoprotein-E. It is also one of the receptors of reelin, an extracellular matrix protein which regulates the processes of neuronal migration and synaptic plasticity. It is associated with parental consanguinity and found in secluded communities such as the Hutterites. Deletion in the VLDLR gene results in cerebellar hypoplasia an autosomal recessive syndrome. VLDLR gene is significantly associated with Reelin pathway.
Physical form
Lyophilized from 0.22 μm filtered solution in PBS, pH 7.4. Normally Mannitol or Trehalose is added as protectants before lyophilization.
Preparation Note
Centrifuge the vial prior to opening. Reconstitute in sterile PBS, pH 7.4 to a concentration of 50 μg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 month. For extended storage, it is recommended to store at -20°C.
