Anti-His tag Antibody, clone 1J13 ZooMAb^® Rabbit Monoclonal

The His-tag is one of the most commonly used purification tags, with six or more consecutive histidine residues. It can be attached to either the N- or the C-terminus of recombinant proteins. Here histidine tends to strongly coordinate with metal ions, such as Ni2+ or Co2+ immobilized on beads or a resin for purification. Since this tag is small, it does not require removal before use of the recombinant protein, and it does not cause any significant interference with the function or structure of a majority of proteins. The His-tagged proteins can be easily detected or purified by used of anti-His-tag antibody. This antibody can also be used to monitor the progress of purification and detection of proteins using immunoblotting, immunostaining, and immunoprecipitation protocols. It can detect proteins with His-tag fused to either the N- or the C-terminus. Quality Control Testing

Evaluated by Western Blotting with His-ABP-tagged recombinant Human Adiponectin.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected His-ABP-tagged recombinant Human Adiponectin.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected His-ABP-tagged, recombinant Human SMCR8 protein.

Affinity Binding Assay: A representative lot of this antibody bound His-tagged, recombinant Human SLC12A4 protein with a KD of 7.8 x 10-6 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected His-tagged AKT1 in HeLa cells transfected to express His-tagged AKT1 protein.

Flow Cytometry (FC) Analysis: 1 µg of this antibody from a representative lot detected His-tagged AKT1 protein in one million HeLa cells.

Clone 1J13 is a ZooMAb^® rabbit recombinant monoclonal antibody that specifically detects His-tagged proteins.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
