General description
Cytotoxic granule associated RNA binding protein TIA1 (UniProt: P31483; also known as Nucleolysin TIA-1 isoform p40, RNA-binding protein TIA-1, T-cell-restricted intracellular antigen-1, p40-TIA-1, TIA-1) is encoded by the TIA1 gene (Gene ID: 7072) in human. TIA1 is a member of a RNA-binding protein family and characterized by its role in post-transcriptional regulation of gene expression, particularly during cellular stress responses. It is a multifunctional protein and possesses nucleolytic activity against cytotoxic lymphocyte (CTL) target cells. It has been suggested that this protein may be involved in the induction of apoptosis as it preferentially recognizes poly(A) homopolymers and induces DNA fragmentation in CTL targets. The major granule-associated species is a 15-kDa protein that is thought to be derived from the carboxyl terminus of the 40-kDa product by proteolytic processing. Structurally, TIA1 contains three RNA recognition motifs (RRMs) and a prion-like domain, which facilitate its interaction with various mRNAs, including those involved in stress granule formation. It is predominantly expressed in heart, small intestine, kidney, liver, lung, skeletal muscle, testes, pancreas, and ovary. TIA1 is localized in the nucleus and accumultaes in cytoplasm, where it localizes to stress granules and processing bodies, acting as a key regulator of mRNA stability and translation, especially under stress conditions such as heat shock or oxidative stress. Alternative splicing resulting in three different isoforms of this gene product has been described in the literature. Dysregulation of TIA1 has been implicated in various diseases, including neurodegenerative disorders like amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD), where its abnormal aggregation is associated with pathological protein inclusions. Additionally, TIA1′s involvement in regulating p53 mRNA localization underscores its critical role in cell cycle control and apoptosis. Application
Evaluated by Western Blotting in U-2 OS cell lysate.
Western Blotting Analysis: A 1:50,000 dilution of this antibody detected TIA1 in U-2 OS cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected TIA1 in lysates from NK-92 cells and Jurkat cells.
Affinity Binding Assay: A representative lot of this antibody bound TIA1 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected TIA1 in Jurkat cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
