General description
Chromobox protein homolog 3 (UniProt: Q13185; also known as HECH, Heterochromatin protein 1 homolog gamma, HP1 gamma, Modifier 2 protein) is encoded by the CBX3 gene (Gene ID: 11335) in human. CBX3 is a major isoform of heterochromatin protein 1 family that recognizes and binds dimethylated and trimethylated histone H3 tails methylated at lysine 9, leading to epigenetic repression. It is reported to promote neuronal maturation, kidney development, differentiation of embryonic stem cells (ESC) to smooth muscle in culture, and embryonic arteriogenesis. Its deregulation has been reported to promote the development of human cancers. It promotes the proliferation, invasion, and tumorigenesis of glioblastoma cells in vitro and in vivo. CBX3 is localized in both euchromatin and heterochromatin (transiently) compartments of interphase nuclei, as well as in the pericentromeric chromatin and arms of mitotic chromosomes. It is phosphorylated by PIM1 during interphase and may be hyper-phosphorylated during mitosis. Specificity
Clone 1K5 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Chromobox protein homolog 3 (CBX3). It targets an epitope within 16 amino acids from the N-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 16 amino acids from the N-terminal half of human Chromobox protein homolog 3 (CBX3).
Application
Quality Control Testing
Evaluated by Western Blotting in Jurkat cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CBX3/HP1 in Jurkat cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CBX3/HP1 in lysates from A549, NIH3T3, and PC12 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CBX3/HP1 in Human testis tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CBX3/HP1 in A431 cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our
reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
