General description

Vacuolar protein sorting-associated protein 4B (UniProt: O75351; also known as EC 3.6.4.6, Cell migration-inducing gene 1 protein, Protein SKD1, Suppressor of K(+) transport growth defect 1) is encoded by the VPS4B (also known as SKD1, SKD1B, VPS4-2, VPS42, MIG1) gene (Gene ID; 9525) in human. SKD1 is a ubiquitously expressed, peripheral membrane protein that is involved in intracellular protein trafficking at the late endosomal protein transport step. Displays about 80% sequence homology with VPS4A. VPS4B has a N-terminal MIT domain (aa 4-82) serves as an adapter for ESCRT-III proteins. It recognizes membrane-associated ESCRT-III (endosomal sorting complex required for transport III) assemblies and catalyzes their disassembly, possibly in combination with membrane fission. Also, redistributes the ESCRT-III components to the cytoplasm for further rounds of multi-vesicular body (MVB) sorting. In conjunction with the ESCRT machinery, it also appears to function in topologically equivalent membrane fission events, such as the terminal stages of cytokinesis and enveloped virus budding (HIV-1 and other lentiviruses). VPS4A/B are required for the exosomal release of SDCBP, CD63 and syndecan. SKD1 is proposed to be monomeric or homo-dimeric in nucleotide-free form and to oligomerize upon binding to ATP to form two stacked hexameric or heptameric rings with a central pore through which ESCRT-III substrates are translocated in an ATP-dependent manner. Application

Quality Control Testing

Evaluated by Western Blotting in HEK-293 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected VPS4B in HEK-293 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected VPS4B in lysates from HepG2 cells and NIH/3T3 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected VPS4B in HEK-293 cells.

Affinity Binding Assay: A representative lot of this antibody bound VPS4B peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 2K14 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects VPS4B. It targets an epitope within 18 amino acids from the N-terminal half.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
