General description

Angiopoietin-like protein 8 (UniProt: Q6UXH0; also known as Lipasin, Refeeding-induced fat and liver protein, Betatrophin) is encoded by the ANGPTL8 (also known as C19orf80, RIFL, UNQ599/PRO1185) gene (Gene ID: 55908) in human. ANGPTL8 is a secreted glycoprotein that plays a critical role in lipid metabolism and energy homeostasis. It promotes pancreatic β-cell proliferation, induces insulin secretion and regulates plasma triglycerides levels. ANGPTL8 contains an N-terminal coiled-coil domain but lacks the C-terminal fibrinogen-like domain. ANGPTL8 forms ANGPTL3/8 and ANGPTL4/8 complexes regulating lipoprotein lipase (LPL) activity, thereby modulating triglyceride levels in circulation and influencing lipid storage in adipose tissue. It is enriched in the liver and adipose tissue, with increased levels observed in response to high-fat diets or insulin signaling. Elevated ANGPTL8 levels have been associated with dyslipidemia, obesity, and type 2 diabetes. ANGPTL8 has been implicated in cardiovascular diseases due to its influence on lipid profiles and inflammation. KLH-conjugated linear peptide corresponding to 24 amino acids from the C-terminal region of human ANGPTL8.

Application

Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ANGPTL8 in HepG2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected ANGPTL8 in lysate from human liver tissue.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected His-tagged, recombinant fragment of human ANGPTL8 .

Affinity Binding Assay: A representative lot of this antibody bound with ANGPTL8 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected ANGPTL8 in rat liver tissue sections.

Immunocytochemistry Analysis (ICC): A 1:100 dilution from a representative lot detected ANGPTL8 in MCF7 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 2K17 is a ZooMAb® rabbit recombinant monoclonal antibody that detects ANGPTL8. It targets an epitope within 24 amino acids from the C-terminal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
