General description
Complement decay-accelerating factor (UniProt: P08174) is encoded by the CD55 (also known as CR, DAF) gene (Gene ID: 1604) in human. CD55 is a cell surface membrane protein associated in lipid microdomains, that acts as a complement regulatory protein. It is composed of four short consensus repeat (SCR) domains, a serine/threonine-rich region, and a glycosylphosphatidylinositol (GPI) anchor that attaches it to the cell membrane. This structure allows it to interact with complement proteins and prevents the formation of the membrane attack complex by dissociating C3/C5 convertases as well as preventing the assembly of C3/C5 convertases. CD55 is widely expressed on the surface of most blood cells, epithelial cells, and endothelial cells. It is also present in various tissues, providing broad protection against complement-mediated damage. It is essential in many organ systems including pregnancy for protection of the semiallogeneic fetus or for preventing uncontrolled infiltration by white cells in their pro- and/or anti-inflammatory functions. CD55 has been implicated in autoimmune disorders, complement hyperactivation, angiopathic thrombosis, and protein-losing enteropathy (CHAPLE), paroxysmal nocturnal hemoglobinuria (PNH), and cancer progression and is overexpressed in several types of cancer. Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CD55 in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CD55 in human lung tissue lysate.
Flow Cytometry Analysis: 1 µg from a representative lot detected CD55 in one million human peripheral blood mononuclear cells (PBMC).
Affinity Binding Assay: A representative lot of this antibody bound with CD55 peptide with at least one thousand-fold (10,000X) higher affinity than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CD55 in human lung tissue sections.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 1J12 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CD55. It targets an epitope within 24 amino acids from the N-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
