General description

ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the N-terminal half of human SRSF1.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected SRSF1 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected SRSF1 in lysates from mouse brain tissue and rat brain tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected SRSF1 in human small intestine tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound SRSF1 peptide with at least ten million-fold (10,000,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Analysis Note

Serine/arginine-rich splicing factor 1 (UniProt: Q07955; also known as Alternative-splicing factor 1, ASF-1, Splicing factor; arginine/serine-rich 1, pre-mRNA-splicing factor SF2, P33 subunit) is encoded by the SRSF1 (also known as ASF, SF2, SF2P33, SFRS1) gene (Gene ID: 6426) in human. It is a member of the highly conserved family of SR proteins, and plays a critical role in both constitutive and alternative pre-mRNA splicing. The protein binds to pre-mRNA transcripts and components of the spliceosome, and can either activate or repress splicing depending on the location of the pre-mRNA binding site. The protein s ability to activate splicing is regulated by phosphorylation and interactions with other splicing factor associated proteins. SRSF1 has an inhibitory role in exon skipping, in order to maintain splicing accuracy and to monitor alternative splicing. SRSF1/ASF also mediates mRNA nuclear export and translation processes and supports genomic stability by impeding formation of mutagenic DNA:RNA hybrid R loop structures between the transcript and the template DNA. Structurally, SRSF1 contains two RNA recognition motifs (RRMs) and a C-terminal serine/arginine-rich (RS) domain. It is ubiquitously expressed and exhibits high levels in immune and proliferative tissues. SRSF1 protein is mostly localized in the nucleus but can also shuttles between the nucleus and the cytoplasm. It is suggested that both the RS domain and an RRM domain are required for nucleocytoplasmic shuttling. Multiple transcript variants encoding different isoforms have been found for this gene. Studies have shown that SRSF1 plays a role in the regulation of caspase-9 alternative splicing as well as being a critical component for ceramide to function as a mediator of alternative splicing for the same caspase. Studies demonstrated that SRSF1 acts as a master switch of gene expression and regulation in our immune system. SRSF1/ ASF is also involved in the replicaton of HIV-1. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
