General description
Every large scale commercial purification begins at a small scale in a method development lab by evaluating retention and selectivity of the target protein versus impurities, host cell proteins, DNA, etc., on multiple resins, often from different vendors. Depending on the constraints, the process development engineer can either purchase bulk resins to pack several small columns with different resins or take advantage of the speed and convenience of pre-packed columns for resin evaluation. Pre-packed ToyoScreen® Process Development Columns are available in 1 mL and 5 mL volumes packed with the most popular Toyopearl® resins. ToyoScreen columns can be used to quickly establish optimum elution conditions. Varying pH, salt type, gradient conditions and flow rate are common experimental parameters explored. Initial results from resin screening and optimization with ToyoScreen columns accurately predict the separation behavior at larger scale.
