Detergents for payload solubilization

Detergents support payload solubilization and impurity clearance studies by Many ADC cytotoxic payloads are highly hydrophobic and require detergent co-solvents for adequate solubility during conjugation and linker-payload preparation. CHAPS, octyl β-D-glucopyranoside, and sodium deoxycholate support homogeneous reaction conditions and improve aggregate solubilization during downstream purification.

Polymers and additives for nonspecific adsorption reduction

At manufacturing scale, nonspecific adsorption of ADC molecules to vessel surfaces, tubing, and filtration equipment causes product loss and yield variability. PEG and poloxamer additives passivate contact surfaces, minimize ADC binding, and support consistent yield recovery, which is critical for cost-effective, reproducible scale-up.

Process buffers for system stability

Buffer composition is a critical process parameter (CPP) that directly influences conjugation efficiency and ADC aggregation at scale. High purity MES, phosphate, and Tris buffers control pH and ionic strength from conjugation through fill-finish.

Redox reagents

Partial reduction of antibody interchain disulfides using TCEP or DTT generates free cysteine thiols for thiol-maleimide conjugation. Precise molar ratio control achieves target DAR values (typically 2–4 or 4–8) and prevents over- or under-reduction, both of which produce heterogeneous product with compromised potency or stability.
