General description

Myc box-dependent-interacting protein 1 (UniProt: O00499; also known as Amphiphysin II; Amphiphysin-like protein; Box-dependent myc-interacting protein 1; Bridging integrator 1, BIN1) is encoded by the BIN1 (also known as AMPHL) gene (Gene ID: 274) in human. BIN1 is expressed in many tissues, but has the highest expression in brain and muscle. In brain it is concentrated in the cytomatrix beneath the plasma membrane of axon initial segments and nodes of Ranvier. In skeletal muscle, it is concentrated around the plasmalemma of T tubules. BIN1 is involved in regulation of synaptic vesicle endocytosis and is also reported to act as a tumor suppressor that inhibits malignant cell transformation. BIN1 is considered as a dominant autoantigen in paraneoplastic Stiff-man syndrome. Mutations in BIN1 gene are known to cause myopathy centronuclear 2 (CNM2), a congenital muscle disorder characterized by progressive muscular weakness and wasting involving mainly limb girdle, trunk, and neck muscles.
