General description

Tomato Juice Broth is specifically designed for the cultivation of yeasts and other aciduric microorganisms. Lactobacilli, which grow poorly on standard culture media, require specialized nutrients for optimal growth. Mickle and Breed demonstrated the effectiveness of tomato juice in selectively inhibiting the growth of non-acidophilic bacteria. Dextrose serves as an additional carbon source, while yeast extract provides nitrogen, carbon, and essential B-complex vitamins. Potassium phosphates function as buffering agents, and the inclusion of several inorganic salts in low concentrations supplies essential ions that, along with yeast extract, support bacterial metabolism.

Tomato Juice Broth is widely used in the food and beverage industry and microbiology research for isolating and cultivating acid-tolerant microorganisms. It is suitable for applications requiring the growth of aciduric bacteria and yeasts, making it ideal for studies related to fermentation and microbial analysis.

Preparation Note

Suspend 41.23 g in 1 litre distilled water. Boil to dissolve the medium completely. Dispense in tubes and sterilize by autoclaving at 121°C for 15 minutes.

The designations basic, plus, or prime are added to indicate the quality control level, from basic quality control to standard QC plus to prime for full regulatory compliance.
