General description

Flotillin 2 encodes a caveolae-associated, integral membrane protein, which is thought to function in neuronal signaling. Caveolae are small domains on the inner cell membrane involved in vesicular trafficking and signal transduction.
Flotillins are lipid raft-associated proteins, and belong to the integral membrane proteins. Flotillins contain an evolutionarily conserved domain called the prohibitin homology (PHB) domain or SPFH domain (stomatin/prohibitin/flotillin/HflK/C domain). The lipid recognition motif present in the PHB domain, is responsible for targeting proteins to the plasma membrane. Flotillin 2 (FLOT2) is widely distributed in human tissues. FLOT2 is mainly present at the plasma membrane, but also localizes in intracellular compartments including lipid droplets and compartments of the endocytic pathway.

Immunogen

synthetic peptide corresponding to amino acids 232-245 of human flotillin 2 with an N-terminal added cysteine, conjugated to KLH. The corresponding sequence is identical in rat and mouse.

Application

Anti-Flotillin 2 antibody is suitable for:
  • indirect immunofluorescence (10-20μg/mL using human HEK-293T cells).
  • western blot (0.5-1μg/mL using whole extract of mouse 3T3 cells).
  • western blot (2-4μg/mL using whole extract of rat PC12 cells).
The antibody has been used for western blotting of proteins isolated from HeLa cells, primary mouse oesophageal epithelial cells and HEK-293 cells.

Biochem/physiol Actions

Flotillins are scaffold proteins which are crucial for the formation and dynamics of lipid rafts. They are associated in different biological processes, such as endocytosis, phagocytosis, neuronal regeneration, insulin signaling etc. FLOT2 (Flotillin 2) is a target of miR-34a. It is up-regulated in nasopharyngeal carcinoma, renal cell carcinoma, breast cancer and gastric cancer. Knockdown of FLOT2 reduces cell proliferation, migration, and invasion. FLOT2 interacts with γ-catenin and regulates cell-cell adhesion in non-malignant epithelial cells.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.
