Human AB serum is used in tissue engineering, transplantation and cell therapy applications for the expansion of mesenchymal stem cells (MSC) from adipose tissue or mesenchymal stromal cells from human bone marrow; for standardized limbal epithelial stem cell graft generation and transplantation; for ex vivo expansion of NK cells from peripheral blood in Hematopoeitic Stem Cell Expansion Medium; and for upgrading pretransplant human islet culture technology.
Human Serum has been used: as a component of phosphate-buffered saline with Tween® detergent (PBST) in bridging enzyme-linked immunosorbent assay (ELISA), as a mock biological sample for assessing the performance of paper-based ELISA assay with complex biological samples, as a diluent to create a dilution series of the baseline sample for the generation of standard curve for NISTmAb spiked into serum, as well as for multiple myeloma (MM) patient sample MM2232

Biochem/physiol Actions

Human AB serum is typically obtained by allowing whole blood to clot naturally, which is known as Off-The-Clot (OTC) serum. Human serum obtained from clotted whole blood is preferred as it usually contains higher levels and a more diverse range of growth factors for optimal cell growth. Human serum from blood-type AB donors lacks antibodies specific for both A and B blood-type antigens. This makes it a universal donor serum that is suitable for various cell culture types including human stem cell lines.

Packaging

Packaging may vary for different batches due to short supply of the bottles used to pack this product. The presence or lack of shrink wrap around the cap of the bottle doesn’t impact product integrity/sterility of the product.

Warning

Each donor is tested for and found non-reactive for Hepatitis B & C and non-reactive for Human Immunodeficiency Virus (HIV) antibody by ELISA. Nevertheless, products of human origin should be considered potentially infectious and handled accordingly.

  1. Which document(s) contains shelf-life or expiration date information for a given product?

    If available for a given product, the recommended re-test date or the expiration date can be found on the Certificate of Analysis.

  2. There is a precipitate in my serum.  Is my serum contaminated?

    Precipitates in serum can often appear as large round bodies, even as doublets per microscopic exam. This can be mistaken as a yeast contamination. Brownian motion may be mistaken for motility. These are lipid aggregates that will be readily dissolved when exposed to lipase. Also, these particles will not gram stain or show any growth in culture on aerobic, anaerobic, or fungal media and culture conditions. The performance of the material is not effected. Centrifugation or filtration of small volumes will remove these particles.

  3. How do I get lot-specific information or a Certificate of Analysis?

    The lot specific COA document can be found by entering the lot number above under the "Documents" section.

  4. Is Product H4522, Human serum, Heat Inactivated (HI)?

    This serum is not heat inactivated. If heat inactivated serum is needed, the serum can be heated to 56 °C for 30 minutes. Heating inactivates the complement cascade. Active complement can participate in cytolytic events, leading to contraction of smooth muscle, histamine release from mast cells and platelets, and activation of lymphocytes and macrophages.

  5. How is Product H4522, Human serum, prepared?

    Blood is collected and has citrate added as an anticoagulant to inhibit clotting.  The blood is then centrifuged and separated into cells and plasma.  The plasma is separated from the cells, and calcium is added to the plasma to activate the clotting cascade.  The preparation is then centrifuged again, and the serum is the liquid product remaining.  This process yields a serum low in growth factors, as it does not contain those factors released from the cells upon clotting.

  6. Product H4522, Human serum, is cloudy. What causes the cloudiness?

    It is not unusual for our human serum products to be cloudy. This product is from pooled serum and cloudiness indicates that lipids are present in the serum. No clear, lipid-free human serum products are available.

  7. Why do we specify male AB in the description of Product H4522, Human serum?

    Male blood is used in serum products, where specified, because female serum can have more antibodies resulting from pregnancy.  Type AB serum is used as there are no anti-A and/or anti-B antibodies that are found in other blood types.

  10. What is the difference between human serum from AB plasma and  human serum from AB clotted whole blood?

    Product H6914 is prepared from whole blood that is allowed to clot, centrifuged and the serum removed. There is no additional processing required.Product H4522 is prepared from anticoagulated (sodium citrate) whole blood. This is centrifuged to pellet the cells (RBC, WBC, some platelets) and the plasma is removed. This plasma is then treated with calcium to activate the clotting cascade. The preparation is centrifuged again and the supernatant is removed. This product can also be called plasma derived serum.Product H6914 will contain more growth factors since it is allowed to spontaneously clot and there will be the release of growth factors from many of the cells (WBC, platelets) present.  The cells are removed before the plasma is clotted for Product H4522, so this plasma will have fewer growth factors present (more similar to what is circulating).
