General description
Dihydropyrimidinase catalyzes the conversion of 5,6-dihydrouracil to 3-ureidopropionate in pyrimidine metabolism. Dihydropyrimidinase is expressed at a high level in liver and kidney as a major 2.5-kb transcript and a minor 3.8-kb transcript. Defects in the DPYS gene are linked to dihydropyrimidinuria. The gene DPYS (dihydropyrimidinase) is mapped to human chromosome 8q22. It is mainly expressed in the liver and kidney cells.

Biochem/physiol Actions
DPYS (dihydropyrimidinase) is an enzyme of the pyrimidine degradation pathway and is responsible for the ring opening of 5,6-dihydrouracil and 5,6-dihydrothymine. Mutations in the DPYS gene can lead to dihydropyrimidinase deficiency, associated with a large accumulation of dihydrouracil and dihydrothymine in urine, blood and cerebrospinal fluid. Methylation pattern in DPYS gene might predict prostate cancer-related mortality.{33)
