General description
Glucuronoxylomannan (GXM) is the principal capsular polysaccharide of Cryptococcus neoformans, a pathogenic fungus causing cryptococcosis, particularly in immunocompromised individuals. Structurally, GXM comprises an -(1→3)-linked mannose backbone substituted with xylose and glucuronic acid residues, and it is O-acetylated at specific mannose units, contributing to its heterogeneity. Synthesized intracellularly, GXM is transported via vesicles to the extracellular space, where it integrates into the fungal capsule and interacts with cell wall components such as glucans and chitin. GXM is critical for fungal virulence, providing protection against phagocytosis, modulating cytokine secretion, and interfering with leukocyte migration. It binds to host receptors, including Toll-like receptors (TLR2, TLR4), CD14, and CD18, facilitating immune evasion. GXM exhibits potent immunosuppressive properties by inhibiting pro-inflammatory cytokines (e.g., IL-1β, TNF- ) while promoting anti-inflammatory cytokines like IL-10. This immunomodulation contributes to disease progression, including cryptococcal meningitis. Additionally, GXM has shown promise as an anti-inflammatory agent in experimental models of septic arthritis, reducing inflammation and cytokine levels. Clone 18B7 is a mouse monoclonal antibody that binds to Glucuronoxylomannan (GXM) from Cryptococcus neoformans.
Physical form
Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
