Specificity

Clone 3J20 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects LKB1/STK11. It targets an epitope within 19 amino acids from the N-terminal half.

Application

Quality Control Testing

Evaluated by Western Blotting in L929 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected LKB1/STK11 in L929 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected LKB1/STK11 in PC12 cell lysates and GST-tagged full length Human LKB1/STK11.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected LKB1/STK11 in Human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound in LKB1/STK11 peptide with a KD of 1.3 x 10-3 in an affinity binding assay.

Enzyme Immunoassay (ELISA) Analysis: Serial of dilutions from a representative lot detected GST-tagged, full-length recombinant Human LKB1/STK11.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Serine/threonine-protein kinase STK11 (UniProt: Q15831; also known as EC:2.7.11.1, Liver kinase B1, LKB1, hLKB1, Renal carcinoma antigen NY-REN-19) is encoded by the STK11 (also known as LKB1, PJS) gene (Gene ID: 6794) in human. LKB1/STK11 is the catalytic component of a trimeric complex composed of LKB1/STK11, STe20 related adaptor (STRAD), and calcium-binding protein 39 (CAB39). This complex tethers LKB1/STK11 in the cytoplasm and stimulates its catalytic activity. It is ubiquitously expressed, and its strongest expression is observed in testis and fetal liver. It serves as a tumor suppressor serine/threonine-protein kinase that controls the activity of AMP-activated protein kinase (AMPK) family members, thereby playing a role in various processes, including cell metabolism, cell polarity, apoptosis, and DNA damage response. It acts by phosphorylating the T-loop of AMPK family proteins, thus promoting their activity. It can also phosphorylate non-AMPK family proteins such as STRADA, PTEN and possibly p53/TP53. It is also reported to regulate cellular polarity by remodeling the actin cytoskeleton and is required for cortical neuron polarization by mediating phosphorylation and activation of BR serine/threonine kinase 1 and 2, leading to axon initiation and specification. LKB1/STK11 is phosphorylated by ATM at threonine 363 following ionizing radiation. Its phosphorylation at serine 428 by ribosomal protein S6 kinase A1 (RPS6KA1) is required to inhibit cell growth. Two isoforms of LKB1/STK11 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
