Specificity
Clone 1O5 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Zinc finger protein 281 (ZNF281). It targets an epitope within 14 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 14 amino acids from the C-terminal region of human Zinc finger protein 281 (ZNF281).
Application
Quality Control Testing
Evaluated by Western Blotting in T47D cell nuclear extract.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected ZNF281 in T47D cell nuclear extract.
Tested Applications
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected ZNF281 in Human gastric cancer tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound ZNF281 peptide with a KD of 8.4 x 10-6 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ZNF281 in A431 cells.

Target description
Zinc finger protein 281 (UniProt: Q9Y2X9; GC-box-binding zinc finger protein 1, Transcription factor ZBP-99, Zinc finger DNA-binding protein 99) is encoded by the ZNF281 (also known as GZP1, ZBP99) gene (Gene ID: 23538) in human. ZNF281 is a transcription factor with four C2H2 type zinc finger domains and plays a KEY® role in controlling cellular stemness. It serves as a transcription repressor and regulates embryonic stem cell (ESC) differentiation. It acts by mediating autorepression of NANOG in ESCs. It binds to the NANOG promoter and promotes association of NANOG protein to its own promoter and recruits the NuRD complex that deacetylates histones. It is reported to repress the transcription of a number of genes including GAST, ODC1 and VIM by binding to the G-rich box in the enhancer region of these genes. ZNF281 is also shown to induce epithelial-mesenchymal transition (EMT) in tumor cells and control the expression of several KEY® EMT-associated genes. Higher expression of ZNF281 is reported in aggressive, undifferentiated stage 4 than in localized stage 1 neuroblastomas and it correlates with poor prognosis in subjects treated with DNA-damaging chemotherapies. The ectopic expression of ZNF281 is shown to inhibit the neuronal differentiation of murine cortical neurons and neuroblastoma cells, and its silencing has an opposite effect. Two isoforms of ZNF281 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity™, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our
reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
